The role of interleukin-22 in mammalian intestinal homeostasis: Friend and foe

  • First Published: 16 February 2024
Description unavailable
Graphical Abstract

Beneficial and harmful effects of IL22 in the intestine. IL-22 is produced by various immune cells at the intestinal epithelium. Here, it acts on enterocytes by binding to the IL-22 receptor complex to initiate its effects. These include inducing cell proliferation/tumorigenesis, wound healing, and induction of mucins by Goblet cells and antimicrobial peptides by Paneth cells. If produced in excessive amounts, these antimicrobial peptides may damage the intestinal epithelium and reduce microbial diversity. IL-22 induces tight junction proteins, modulating epithelial barrier permeability. IL-22 also induces Paneth cells differentiation in humans (image created with Biorender).

MiR-9-enriched mesenchymal stem cells derived exosomes prevent cystitis-induced bladder pain via suppressing TLR4/NLRP3 pathway in interstitial cystitis mice

  • First Published: 02 February 2024
Description unavailable

This study demonstrated that extracellular vesicles derived from miR-9-enriched mesenchymal stem cells can freely cross the blood-brain barrier into the spinal cord tissue, and then target TLR4 receptors on the activated spinal cord glial cell membrane of interstitial cystitis/bladder pain syndrome (IC/BPS) model to inhibit the activation of NLRP3 inflammatorome and release of inflammatory factors, which can reduce neuron stimulation and restore neuron firing threshold, so as to desensitize neurons and achieve the purpose of treating bladder pain in IC/BPS patients.

Dexmedetomidine activates the PKA/CREB pathway and inhibits proinflammatory factor expression through β2 adrenergic receptors

  • First Published: 27 February 2024
Description unavailable

The inflammatory cell model of human mononuclear macrophage (THP-1) cells induced by lipopolysaccharide (LPS) was established to study the effect of Dexmedetomidine (DEX) on the expression of cell-related inflammatory factors. ADRA2A gene knockout THP-1 cells (THP-1KO) were constructed, and the effect of DEX on the expression of inflammatory factors of THP-1KO cells was detected. The target sites of DEX on β2 AR were screened by molecular docking. Reversion experiments were performed using ADRB2siRNA. The results showed that DEX significantly decreased the expression of MCP-1, interleukin-06, and IL-8 in THP-1 and THP-1KO cells induced by the LPS, and increased the level of IL-10. Immunofluorescence results showed that β2 adrenergic receptor (β2 AR) expression increased after DEX treatment. DEX significantly increased the activation of β2 AR and PKA/CREB pathways, inhibited the activation of NF-κB. ADRB2 siRNA reversed the effects of DEX and promoted the release of inflammatory cytokines

Galectin-9 alleviates acute graft-versus-host disease after haplo-hematopoietic stem cell transplantation by regulating regulatory T cell/effector T cell imbalance

  • First Published: 14 February 2024
Description unavailable

In patients with acute graft-versus-host disease (aGVHD), the expression of Tim-3 is significantly increased. Galectin-9 binding to Tim-3 may inhibit the activation of the PI3K/AKT pathway and enhance the function of Treg cells. On the other hand, transforming growth factor (TGF)-β promotes the differentiation of Treg cells through autocrine secretion, while TGF-β induces the expression of Galectin-9 in a paracrine manner. The increased Treg cells can inhibit the activation of Th1 and Th17 cells by secreting TGF-β, thus alleviating aGVHD and inducing immune tolerance

Our study highlights the usefulness of the C-reactive protein (CRP) to prognostic nutritional index (PNI) ratio in predicting the in-hospital mortality of severe fever with thrombocytopenia syndrome (SFTS) patients based on a cohort of 610 SFTS cases from Wuhan Union Hospital. Subsequent survival analysis demonstrates that CRP/PNI could well differentiate SFTS individuals with high risk from those with low risk. receiver operating characteristic (ROC) analysis unveils that CRP/PNI possesses nice predictive power for predicting the in-hospital mortality of SFTS. Briefly, CRP/PNI is a reliable biomarker that can serve as a promising predictor for subjects with SFTS.
